Single or small number of FACS sorted lymphocyte in 96-well plates (i.e., up to ~500 cells/sample)
Prior to FACS, prepare 96-well plates with pre-aliquoted 5 µl of deionized water (or PBS) per well. This ensures the cells are suspended in the liquid and prevents the cell from getting stuck to the tube surface and dehydrating. Immediately after sorting, remove the plate from the motorized stage and seal it to prevent desiccation (nucleic acids can be absorbed by the plastic plate if it gets too dry). Then, briefly centrifuge the plate for 10-15 sec to propel any drops on the side walls into the water, label, freeze and store the plate at -80°C, and ship it to Cellecta on dry ice. Be sure to put the plates in a solid cardboard box or other case to prevent detaching the seal and damage from dry ice. Provide information indicating what samples are in each plate and what wells were used for sorting.
DirectCell AIR assay for 500-50,000 purified lymphocyte cell populations.
Collect cells in 1xPBS or any commonly used buffer without magnesium. Transfer the cells into 1.5-ml or 0.5-ml test tubes, spin down at low speed (e.g. 500g for 5 min) and remove excess supernatant from cell pellet keeping residual volume in test tube 15-25 µl (to prevent loss of cells due to complete removal of supernatant). Put the test tubes with cell pellets in a cardboard box, freeze, store at -80°C, and ship on dry ice. Clearly label each tube with numbers (from 1 to X), and a short name (at most, 6-8 printed alphanumeric characters), and send us the Sample Submission Form with the sample names.
AIR assay for large (more than 50,000 lymphocytes) cell/tissue samples.
For larger cell samples with more than 50,000 lymphocyte cells, we will purify RNA and DNA and perform AIR assay. Transfer the cells into 1.5-ml test tubes, spin down at low speed (e.g. 500g for 5 min) and remove excess supernatant from cell pellet. Add 0.35-ml of RTL-Plus Buffer (Qiagen Cat. No.1053393), freeze -* at -80°C, and ship to Cellecta on dry ice. Clearly label each tube with numerical numbers (from 1 to X), and short name (at most, 6-8 printed alphanumeric characters), and send us the Sample Submission Form with the sample names.
