To validate the expression of a control TCR construct encoding specificity for EBV, CMV, and FLU peptides in Jurkat Reporter Cells, we performed surface staining using FLU, CMV, and EBV peptide-specific dextramers (Immudex Reagents – Cat# WB02132, WB02130, WB02161). Jurkat-TCR cells were stained with the following dextramers. Stained cells were then analyzed by flow cytometry to confirm surface expression and binding specificity (see Figure 4).

Fig 4. Map of Jurkat TCR Knockout (K/O) NFAT-GFP Reporter Cell Line The schematic shows the Jurkat TCR K/O NFAT-GFP reporter line transduced with a control TCR construct targeting EBV, FLU, or CMV peptides. Fluorescently labeled dextramers specific for each peptide bind to the corresponding TCRs expressed on the surface of the Jurkat reporter cells and are detected via FACS analysis.

Fig 5. Jurkat Reporter Clone staining with peptide-specific dextramer (CMV, FLU and EBV). Results show positive staining of Jurkat clone expressing peptide specific TCR.
